In contrast to traditional dyes, the Brilliant Violent fluorescent polymers are about 80 kD in size, consisting of fluorescent monomeric subunits that act cooperatively along the entire length of the polymer backbone. The energy is conducted much like a molecular antennae to capture light and pass it down the antennae like a lightning rod. This creates an extinction coefficient that is substantially larger than standard organic fluorescence, and when conjugated to antibodies, this results in high intensity brightness on labeled cells.

These conjugated polymers are comprised of alternating single and double bonds and aromatic units. This repeating bond structure creates a continuous π-orbital system and extended electronic delocalization. A configuration that confers several unique and tunable optical properties, (including large extinction coefficients (>106 M-1cm-1), intense photoluminescence, and massive collective response); that collectively address fundamental limitations in fluorescence detection sensitivity.

Current violet excited dyes used in flow cytometry, like Pacific Blue™ and BD Horizon V450, have high quantum yields but limited extinction coefficients. Because the extinction coefficient of a conjugated polymer is directly proportional to the degree of polymerization (or number of repeat units), these structures are designed for improved brightness. Further, as these polymers are derived from common synthetic organic and polymer chemistry techniques is it possible to manufacture reagents very reproducibly; in terms of size, conjugation sites, and optical properties.

In addition, unlike quantum dots, these new conjugated polymers have discrete excitation spectra, similar to that of organic dyes, which minimizes potential issues with cross-beam compensation.

BV421™ has both a narrow excitation curve and a narrow emission curve, which means less compensation than other fluorochromes.

BV421™ enables resolution of antigens with directly conjugated antibodies, providing increased sensitivity for resolving weakly expressing antigens and expanding options for multi-color panels.
